  • PTHR ID & PTN node:
    CHLORIDE CHANNEL PROTEIN (PTHR45711) PTN000189250

  • Sequences with problematic annotation (ID + gene/protein name):

  • FBgn0036566 ClC-c is getting voltage-gated chloride channel activity (GO:0005247) and plasma membrane (GO:0005886) from PTN000189250

  • Type of Issue: Erroneous source or erroneous propagation, or other issue

  • This might be due to the way the tree is split.

  • The ClC family are split into two distinct mechanistic groups
    From PMID:29845874:
In mammals, ClC-1, -2, -Ka/-Kb are plasma membrane Cl- channels, whereas ClC-3 through ClC-7 are 2Cl-/H+-exchangers in endolysosomal membranes.
    D.mel -> human orthologs - mech
    FBgn0051116 ClC-a - CLCN2 - voltage-gated Cl- channel
    FBgn0033755 ClC-b CLCN7 - Cl-/H+ antiporter
    FBgn0036566 ClC-c CLCN3 - Cl-/H+ antiporter

FBgn0051116 ClC-a
Annotations are good:
voltage-gated chloride channel activity PANTHER:PTN000189253
plasma membrane PANTHER:PTN000189253

FBgn0033755 ClC-b
Annotations are good:
chloride transmembrane transporter activity PANTHER:PTN001363397 (even better if it were chloride:proton antiporter activity)
intracellular membrane-bounded organelle PANTHER:PTN001363397
lysosomal membrane PANTHER:PTN002481857

FBgn0036566 ClC-c
Incorrect:
voltage-gated chloride channel activity PANTHER:PTN002269368

Have a really mixed bag for CC for ClC-c that doesn't make sense:
early endosome PANTHER:PTN002269368
plasma membrane PANTHER:PTN000189250
synaptic vesicle PANTHER:PTN002481889

we just have it characterised in early/recycling endosome
