Extracting particles into a stack

When it comes to extracting particles into a stack of particles, there are two broad choices for the approach required in order to extract the particles:

  1. Extracting particles outside of Relion
  2. Extracting particles using Relion

When should I use Relion for particle extraction?

In principle, you can use Relion for all types of data processing - ranging from negative stain to high-resolution cryo-EM.

In practice, however, Relion's CTF correction routine may not be required for negative stain image analysis. In this case, you might consider extracting particles outside of Relion, or you could extract particles in Relion withOUT CTF correction.

Relion is designed for high-resolution cryo-EM structure determination
